I am planning a gene synthesis experiment.
QUESTIONS:

1) Is there new knowledge on this topic, a new powerful protocol? Otherwise I would like to assemble the gene from overlapping oligonucleotides covering both strands completely, maybe followed by PCR-amplification... any suggestions?

2) Is there a public domain software which helps to split down the DNA sequence to the level of oligonucleotides, helping in avoiding self-complementary oligonucleotides and checking for repeats or other homologous regions that could interfere with correct assembly of the gene?
