I am facing problems with PCR amplification of genomic DNA isolated from amniotic fluid. Can anybody share their experience and trouble shooting for the same. Is it due to some inhibitors or any other reason. Some such DNA samples do amplify after treatment with resins. However, I would like to know about a protocol for isolation of DNA from Amniotic fluid which does not lead to such problems

May be able to help troubleshoot. Have 11 years experience studying the minutiae of PCR and its kinetics. Protocols are dated, but might be useful.
