In vitro gene-expression

Program:
The subject is recognition and function af transcription-factors at DNA promoter-regions in front of genes. The course contains both a theoretical and a practical part.
Gene-expression will functionally be illustrated by in vitro transcription and translation using purified proteins and cell extracts. Mechanisms of both procaryotic RNA polymerase and eukaryotic RNA polymerase II and III will be examined.
In addition DNA recognition of promoter-regions will be studied by gel-shift assays and enzymatic DNase I, chemical DMS and KMnO4 and photochemical uranyl footprinting techniques.
Finally Peptide Nucleic Acids (PNA) will be used as an illustrating example of how the expression of specific genes can be blocked.
